Drug intelligence / Profile preview
Casdatifan plus ivonescimab is an investigational combination regimen being evaluated for the treatment of clear cell renal cell carcinoma (ccRCC), including first-line metastatic disease. Casdatifan (AB521) is an oral, once-daily small-molecule inhibitor of hypoxia-inducible factor 2-alpha (HIF-2α) developed by Arcus Biosciences. Ivonescimab (SMT112/AK112) is an investigational humanized tetravalent bispecific antibody developed by Akeso and licensed to Summit Therapeutics, which simultaneously targets programmed cell death protein 1 (PD-1) and vascular endothelial growth factor (VEGF). Under a clinical trial collaboration announced in 2026, Arcus sponsors and conducts the combination study as a new cohort within its ARC-20 platform trial, with Summit supplying ivonescimab. The combination aims to provide a well-tolerated, tyrosine kinase inhibitor (TKI)-sparing treatment option for patients with advanced kidney cancer.
