casdozokitug + toripalimab

**casdozokitug + toripalimab** is an experimental combination therapy currently under clinical investigation for unresectable, locally advanced, or metastatic hepatocellular carcinoma (HCC). Casdozokitug is a first-in-class monoclonal antibody targeting interleukin-27 (IL-27), designed to reverse IL-27-mediated immune suppression by enhancing IFN-γ and NK cell activation. Toripalimab is a monoclonal antibody targeting the programmed death receptor-1 (PD-1), working as an immune checkpoint inhibitor to enhance T cell-mediated antitumor responses. This combination is being tested with/without other agents (notably bevacizumab) for synergistic immunomodulatory and antitumor effects. Both drugs are biologics, with clinical trial sponsorship and development by Coherus BioSciences. As of September 2025, this combination has not received FDA approval.
