Elgia Therapeutics' caspase-1 inhibitor is a preclinical small-molecule drug candidate designed to treat chronic inflammatory and fibrotic diseases. It functions by selectively inhibiting caspase-1, a cysteine protease that plays a critical role in the NLRP3 inflammasome pathway. By blocking caspase-1 activity, the drug prevents the proteolytic cleavage and activation of pro-inflammatory cytokines, specifically interleukin-1 beta (IL-1β) and interleukin-18 (IL-18), thereby suppressing downstream inflammatory cascades. The program is currently being evaluated for the treatment of cryopyrin-associated periodic syndrome (CAPS) and inflammatory bowel disease (IBD), including colitis.
