Elgia Therapeutics is developing small-molecule inhibitors of caspase-6, an enzyme critical in the pathways of inflammation, programmed cell death (apoptosis), and fibrosis. These inhibitors are designed to be potent and selective, targeting both the zymogen (inactive) and active forms of the enzyme with IC50 values in the 1-10 nM range. By inhibiting caspase-6, the therapy aims to halt inflammatory signaling and tissue scarring. The program is currently in preclinical development for the treatment of fibrotic diseases (including kidney disease), neuroinflammation (with potential applications in Alzheimer's disease), and non-alcoholic steatohepatitis (NASH). Preclinical models have demonstrated efficacy in high-fat diet mouse models of NASH and protection of neurons from inflammation in caspase-6 knockout models.
