CATP LNP + SamRNA-IL12 is an experimental cancer immunotherapy that utilizes Cascade Amplification of Therapeutic Payloads (CATP) technology. This approach employs lipid nanoparticles (LNPs) to co-deliver self-amplifying mRNA (SamRNA) encoding the immunostimulatory cytokine interleukin-12 (IL-12) along with modified mRNA encoding alphavirus capsid and envelope proteins. The therapy functions through a dual-amplification mechanism: the SamRNA replicates within the initially transfected cells to drive high levels of IL-12 expression, while the structural proteins package the SamRNA into defective viral particles (DVPs). These DVPs are then released to transduce neighboring cells, effectively propagating the therapeutic payload throughout the tumor microenvironment. Preclinical studies have demonstrated efficacy in models of B16F10 melanoma and pancreatic ductal adenocarcinoma by enhancing anti-tumor immune responses.
