Cavrotolimod + cemiplimab is an investigational combination therapy being studied for advanced solid tumors, especially cutaneous squamous cell carcinoma (CSCC) and Merkel cell carcinoma (MCC) resistant to prior PD-1/PD-L1 blockade. Cavrotolimod is a spherical nucleic acid configuration of a toll-like receptor 9 (TLR9) agonist oligonucleotide that stimulates both innate and adaptive immune responses against tumors. Cemiplimab is a monoclonal antibody targeting programmed cell death protein 1 (PD-1), blocking its inhibitory effects on T-cells and promoting antitumor immune activity. The combination is intended to trigger and sustain antitumor immune responses in tumors refractory to PD-1 pathway blockade, and has shown early safety and potential efficacy in phase 1b/2 trials[1][2][3][4].
