Overview

**CC-122 (avadomide) + rifampin** is an experimental **combination therapy** that has not been widely reported in clinical development as a fixed combination, but both drugs are individually well described. CC-122 (avadomide) is a *cereblon E3 ligase modulator (CELMoD)*, related to the IMiD class (immunomodulatory imide drugs). It binds cereblon in the cullin 4 E3 ubiquitin ligase complex, inducing degradation of the transcription factors Aiolos and Ikaros. This leads to T-cell activation, apoptosis of malignant B cells, and immunomodulatory effects, with primary investigation in non-Hodgkin lymphoma and other cancers. Rifampin is a well-known broad-spectrum *antibiotic* of the rifamycin class, most commonly used to treat tuberculosis and sometimes as a pharmacological probe for CYP3A induction in drug-drug interaction studies. No evidence suggests this combination is used as a marketed or established therapy, but it could be studied to assess pharmacokinetics of CC-122 in the presence of a strong CYP3A4 inducer (rifampin), as rifampin is a standard agent for such drug-drug interaction studies.
