Overview

CC-486 + pembrolizumab is a combination therapy consisting of oral azacitidine (CC-486), an epigenetic modulator and DNA methyltransferase inhibitor, and pembrolizumab, a monoclonal antibody immune checkpoint inhibitor targeting programmed cell death protein 1 (PD-1). This combination is being investigated for its potential to enhance antitumor immune responses by combining the immunomodulatory effects of hypomethylating agents with the immune activation provided by PD-1 blockade. The primary indications under investigation include advanced non-small cell lung cancer (NSCLC), platinum-resistant epithelial ovarian cancer, fallopian tube or peritoneal cancer, and melanoma[1][2][3][4]. Clinical trials have evaluated safety and efficacy in these populations; however, no significant improvement in progression-free survival was observed in NSCLC compared to standard therapy[1].
