Overview

**CCS1477 + atezolizumab** is an investigational combination therapy consisting of CCS1477 (inobrodib), an oral small molecule inhibitor of the p300/CBP bromodomain, and atezolizumab, a monoclonal antibody targeting PD-L1 (programmed death-ligand 1). CCS1477 inhibits the activity of transcriptional co-activators p300/CBP, disrupting critical pathways in cancer cell growth and resistance mechanisms, particularly androgen receptor (AR) signaling, and modulating immune evasion in the tumor microenvironment. Atezolizumab blocks the interaction between PD-L1 and its receptors (PD-1 and B7.1), enhancing T-cell effector function and promoting anti-tumor immunity. This combination is being evaluated primarily in patients with non-small cell lung cancer (NSCLC) who have progressed after chemotherapy, aiming to overcome resistance to immunotherapy and deliver enhanced anti-tumor efficacy through dual epigenetic and immune checkpoint inhibition[1][2][3][4].
