Overview

CD137+ VETc (vaccine-educated T cells enriched by CD137 expression) is an experimental autologous adoptive cell therapy being investigated for the treatment of Acute Myeloid Leukemia (AML). The therapy is produced by co-culturing patient-derived T cells with a dendritic cell/AML fusion vaccine in the presence of cytokines (IL-2, IL-7, and IL-15) to generate vaccine-educated T cells (VETc). These cells are then further selected using a biotinylated agonistic antibody against CD137 (4-1BB), a co-stimulatory receptor that serves as a marker for activated, antigen-specific effector cells. The enriched CD137+ population is expanded via CD3/CD28 stimulation to produce a final product with enhanced anti-tumor activity and a memory phenotype. Preclinical studies have shown that this enrichment process significantly improves the cytotoxicity and persistence of the T cells against AML models compared to non-enriched controls.
