CD146-targeted CAR T cells + HER2-targeted CAR T cells is an investigational sequential cell therapy regimen developed by Essen Biotech for the treatment of advanced, relapsed, or refractory sarcomas. The treatment involves the administration of two distinct autologous CAR-T cell products: one targeting the Melanoma Cell Adhesion Molecule (CD146/MCAM) and another targeting the Human Epidermal Growth Factor Receptor 2 (HER2/ERBB2). CD146 is a cell surface glycoprotein associated with tumor metastasis and angiogenesis in sarcomas, while HER2 is a well-characterized tyrosine kinase receptor often overexpressed in various solid tumors. By sequentially targeting these two antigens, the therapy aims to enhance anti-tumor efficacy and mitigate the risk of immune escape in the heterogeneous tumor microenvironment of sarcomas.
