CD19 + CD22 bispecific CAR T-cell therapy is an investigational chimeric antigen receptor (CAR) T-cell therapy designed to simultaneously target two B-cell surface antigens, CD19 and CD22. This dual-targeting approach aims to reduce the risk of relapse caused by antigen escape, a common resistance mechanism in single-target CD19 CAR-T therapies where leukemia or lymphoma cells lose CD19 expression. The therapy involves genetically modifying a patient's own T-cells to express a bispecific CAR construct, typically incorporating single-chain variable fragments (scFvs) for both targets linked to intracellular signaling domains like CD3ζ and costimulatory domains such as 4-1BB. It is primarily being evaluated for relapsed or refractory B-cell malignancies, including B-cell acute lymphoblastic leukemia (B-ALL) and large B-cell lymphoma (LBCL).
