CD19 + CD22 CAR T-cell therapy is an investigational autologous cellular immunotherapy designed to treat B-cell malignancies by simultaneously targeting two surface antigens, CD19 and CD22. This dual-targeting approach aims to mitigate the risk of relapse caused by antigen escape, a common resistance mechanism where cancer cells lose or downregulate CD19 expression following treatment with monospecific CD19-targeted CAR-T therapies. The therapy involves genetically engineering a patient's own T-cells to express chimeric antigen receptors (CARs) through various molecular strategies, including tandem constructs (incorporating both binding domains into one receptor), bicistronic vectors, or the co-administration of two separate CAR-T products. It is primarily being evaluated for relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL) and various forms of B-cell non-Hodgkin lymphoma (B-NHL), including large B-cell lymphoma (LBCL).
