CD19 CAR-T + CD19 CAR-DC is a combination cell therapy comprising two distinct autologous cellular products engineered to target the B-cell antigen CD19. The first component, CD19 CAR-T, consists of T lymphocytes genetically modified to express a chimeric antigen receptor (CAR) specific for the surface protein CD19, which is highly expressed on B-cell malignancies. These engineered T cells recognize and kill malignant and normal B cells expressing CD19 through MHC-unrestricted mechanisms involving cytolytic granule release (perforin/granzyme), Fas/FasL-mediated apoptosis, and cytokine secretion[2][4][6]. The second component, CD19 CAR-DC, refers to dendritic cells (DCs) that are also genetically modified with a similar or identical anti-CD19 chimeric antigen receptor. While less common than T cell-based approaches, dendritic cell-based therapies can enhance antitumor immunity by presenting tumor antigens more effectively and activating endogenous immune responses[6]. This dual approach aims to leverage both direct cytotoxicity from the T cells and enhanced immune activation from the dendritic cells for improved efficacy against relapsed or refractory B-cell malignancies.
