Overview

CD19 CAR-T cells (Xinqiao Hospital) is an autologous chimeric antigen receptor (CAR) T-cell therapy developed by Xinqiao Hospital of Chongqing for the treatment of relapsed or refractory CD19-positive B-cell malignancies, including diffuse large B-cell lymphoma (DLBCL), follicular lymphoma (FL), and mantle cell lymphoma (MCL). The therapy consists of patient-derived T cells transduced with a retroviral vector to express a second-generation CAR comprising an anti-CD19 single-chain variable fragment (scFv) linked to a CD28 costimulatory domain and a CD3-zeta signaling domain. A key feature of this specific program is the investigation of cytokine programming during ex vivo expansion; the "MeCAR" study compares traditional IL-2 expansion with an IL-7/IL-15-based protocol designed to enrich for memory T-cell phenotypes (Tcm/Tn), potentially enhancing in vivo persistence and anti-tumor efficacy.
