Overview

This is an autologous, second-generation chimeric antigen receptor (CAR) T-cell therapy targeting the CD19 antigen, developed by the M.D. Anderson Cancer Center. Unlike many CAR T therapies that use viral vectors, this therapy utilizes the Sleeping Beauty (SB) non-viral gene transfer system (a transposon/transposase system) to introduce the CAR transgene into T cells. The CAR construct includes a CD19-specific single-chain variable fragment (scFv) linked to CD28 co-stimulatory and CD3-ζ signaling domains. It is primarily investigated for patients with high-risk or advanced B-cell lymphoid malignancies, often in the context of autologous hematopoietic stem cell transplantation to eliminate residual disease.
