This therapeutic regimen, developed by the H. Lee Moffitt Cancer Center and Research Institute, combines CD19-targeted Chimeric Antigen Receptor (CAR) T-cell therapy with the 13-valent pneumococcal conjugate vaccine (PCV13). The CAR T-cell component is a personalized immunotherapy where a patient's T cells are genetically engineered to target the CD19 protein on B cells, effectively treating refractory B-cell lymphomas but also causing prolonged B-cell aplasia. To counter the resulting increased risk of bacterial infections, the PCV13 vaccine is administered before and after the CAR T-cell infusion to optimize cellular and humoral immunity against Streptococcus pneumoniae. This integrated approach is currently being evaluated in a Phase 2 clinical trial to improve outcomes and safety for patients with relapsed or refractory B-cell lymphomas.
