CD19-targeted CAR-T cells (Jiuwei Cui) is an investigator-initiated, second-generation chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of CD19-positive B-cell malignancies, specifically recurrent or refractory B-cell acute lymphoblastic leukemia (B-ALL). The therapy involves the genetic modification of a patient's own T cells using a lentiviral vector to express a chimeric antigen receptor consisting of an anti-CD19 single-chain variable fragment (scFv) linked to intracellular signaling domains, typically including a costimulatory domain (such as 4-1BB or CD28) and the CD3-zeta signaling chain. Upon re-infusion into the patient, these engineered T cells recognize and bind to the CD19 antigen expressed on the surface of malignant B cells, leading to T-cell activation, proliferation, and targeted cytotoxicity. This specific program is led by investigator Jiuwei Cui at the First Hospital of Jilin University.
