CD3ζ:MyD88 fusion protein is a recombinant signaling construct designed for use in adoptive cell therapy (ACT), specifically for engineering tumor-infiltrating lymphocytes (TILs). The protein consists of the signaling domain of the CD3ζ chain, a critical component of the T-cell receptor (TCR) complex, fused to MyD88, a key adapter protein in the Toll-like receptor (TLR) and interleukin-1 receptor (IL-1R) pathways. This fusion allows for the activation of MyD88-mediated costimulatory signals in a TCR-dependent manner upon antigen recognition. Developed by researchers at the University of Colorado Anschutz Medical Campus, the construct is intended to enhance T-cell expansion, persistence, and polyfunctionality while reducing exhaustion, thereby improving the efficacy of T-cell therapies against solid tumors such as melanoma.
