Overview

CD33.CAR(2A)CXCR4-CIK cells is an experimental adoptive cell therapy consisting of cytokine-induced killer (CIK) cells engineered to co-express a chimeric antigen receptor (CAR) targeting CD33 and the chemokine receptor CXCR4. Developed primarily by researchers at the Tettamanti Research Center and the University of Milano-Bicocca, this therapy utilizes a bicistronic Sleeping Beauty transposon vector where the two components are linked by a 2A peptide. The CD33-targeting CAR provides antigen-specific cytotoxicity against CD33-positive acute myeloid leukemia (AML) blasts. The overexpression of CXCR4 is intended to restore bone marrow homing capabilities that are typically lost during ex vivo CIK cell expansion, thereby enhancing the eradication of leukemic stem cells within the bone marrow niche.
