Overview

CD34+ bone marrow stem cells are a population of multipotent hematopoietic progenitor and stem cells isolated from human bone marrow based on the expression of the CD34 surface glycoprotein. These cells are capable of self-renewal and differentiation into all blood cell lineages, including erythrocytes, leukocytes, and platelets. CD34 is used as a marker to identify and enrich these primitive blood- and bone-marrow-derived progenitor populations for both research and clinical applications. Clinically, CD34+ bone marrow stem cell transplantation is used to reconstitute the hematopoietic system after chemotherapy or radiation in malignant (e.g., leukemia) or non-malignant (e.g., sickle cell disease) conditions. More recently, autologous intravitreal injection of these cells has been investigated as a regenerative therapy for retinal degenerative diseases such as retinitis pigmentosa; early-phase trials have demonstrated safety and potential efficacy in vision improvement[6][8][10]. The mechanism involves homing to damaged tissues with possible paracrine support for tissue repair[6]. These therapies use either freshly isolated or ex vivo expanded CD34+ HSCs obtained via immunomagnetic selection from donor or patient-derived bone marrow.
