CD34+ enriched peripheral blood hematopoietic stem cells (PBSC) is an investigational cellular therapy developed by Northwestern University for the treatment of hematologic malignancies. This therapy involves the isolation and enrichment of hematopoietic stem and progenitor cells expressing the CD34 surface marker from a donor's peripheral blood using immunomagnetic selection. In the clinical trial context, these enriched cells are used to augment unmanipulated bone marrow in allogeneic transplantation. The mechanism of action relies on the ability of these stem cells to home to the recipient's bone marrow and differentiate into all hematopoietic lineages, thereby reconstituting the blood and immune systems following myeloablative conditioning. The enrichment of CD34+ cells is intended to optimize the graft composition to improve engraftment kinetics and potentially modulate the risk of graft-versus-host disease.
