Overview

CD34+ selected bone marrow mononuclear cells are a population of human hematopoietic stem and progenitor cells isolated from the mononuclear fraction of bone marrow using positive selection for the CD34 surface antigen. CD34 is a transmembrane glycoprotein that serves as a marker for primitive blood- and bone marrow-derived progenitor cells, especially hematopoietic and endothelial stem cells. These multipotent CD34+ stem/progenitor cells can differentiate into all hematopoietic lineages (myeloid and lymphoid) and are capable of self-renewal. Clinically, they are used in autologous or allogeneic transplantation to reconstitute the hematopoietic system after myeloablative therapy in malignant (e.g., leukemia, lymphoma) and non-malignant diseases (e.g., sickle cell disease). They have also been investigated for regenerative applications such as spinal cord injury, liver cirrhosis, peripheral vascular disease, myocardial ischemia, stroke recovery, burns, osteoarthritis, type 1 diabetes mellitus, Parkinson’s disease and other conditions. The mechanism of action involves engraftment in the recipient’s bone marrow niche with subsequent multilineage differentiation to restore or augment blood cell production; additional effects may include paracrine support for tissue repair via angiogenesis[1][3][4][5][6].
