Overview

CD34+ selected hematopoietic stem cells are an investigational cell therapy product developed at Stanford University for use in haploidentical hematopoietic stem cell transplantation (HSCT). The therapy involves the positive selection of CD34+ progenitor cells from a half-matched (haploidentical) donor, which effectively depletes the graft of T-cells to minimize the risk of graft-versus-host disease (GvHD). This approach is specifically evaluated in older or medically unfit patients with various hematologic malignancies, including non-Hodgkin lymphoma, chronic lymphocytic leukemia, and acute myeloid leukemia, as well as severe aplastic anemia. The transplantation protocol utilizes a non-myeloablative conditioning regimen consisting of total lymphoid irradiation (TLI) and anti-thymocyte globulin (ATG), designed to promote stable donor engraftment and a graft-versus-tumor effect with reduced toxicity compared to traditional myeloablative regimens.
