A CD34+ stem cell boost is a therapeutic cell therapy procedure involving the infusion of additional hematopoietic stem cells (HSCs) into a patient who has previously undergone an allogeneic hematopoietic cell transplant (HCT) but is experiencing poor graft function or graft failure. The cells are typically enriched for the CD34+ surface marker, which identifies hematopoietic progenitor cells. This boost is often administered without prior conditioning (chemotherapy or radiation), a strategy known as an unconditioned boost, to enhance donor chimerism and improve blood counts without the toxicity associated with a full second transplant. It is primarily utilized as a salvage option in the context of hematologic disorders such as sickle cell disease or following HCT for various malignancies.
