CD34+ stem cell-derived natural killer cells is an autologous adoptive cell therapy being developed by Hiroshima University and the Japan Agency for Medical Research and Development (AMED). The therapeutic process involves mobilizing a patient's peripheral blood CD34+ hematopoietic stem cells using granulocyte colony-stimulating factor (G-CSF), followed by apheresis and ex vivo differentiation into mature natural killer (NK) cells. These cells are then re-infused intravenously into the patient. The therapy is primarily indicated for the prevention of recurrence in patients with high-risk hepatocellular carcinoma (HCC) who have undergone curative hepatectomy. The mechanism of action relies on the innate ability of NK cells to recognize and eliminate residual malignant cells through direct cytotoxicity, potentially overcoming the immunosuppressive tumor microenvironment.
