CD4 CAR+CCR5 ZFN T-cells are a cell therapy approach for HIV treatment that combines two technologies: chimeric antigen receptor (CAR) T-cell therapy and zinc finger nuclease (ZFN) gene editing. This therapy involves modifying a patient's own CD4+ T cells to express a CD4-based chimeric antigen receptor that can recognize and bind to HIV envelope glycoproteins on infected cells, while simultaneously using ZFN technology to disrupt the CCR5 gene in these cells. The CCR5 gene encodes the main coreceptor for HIV entry, so its knockout confers resistance to HIV infection, protecting the engineered cells. These modified T cells can directly control HIV replication, augment HIV-specific immune responses, and potentially contribute to HIV cure strategies by targeting the latent viral reservoir.
