CD40L + IL2 is an investigational combination therapy most commonly implemented as a vaccine or cell-based immunotherapy, where CD40 ligand (CD40L) is used to enhance antigen presentation and T cell activation, and interleukin-2 (IL2) provides proliferative and activating signals to T cells. This synergistic combination promotes robust antitumor immune responses, increasing MHC-restricted cytotoxic and helper T cell populations and, in some patients, antibody responses to tumors or malignant cells. It has been used in clinical trials for hematological malignancies (e.g., acute myeloid leukemia, chronic lymphocytic leukemia) and solid tumors (e.g., melanoma), often through autologous tumor vaccine platforms, gene-modified cell vaccines, or adoptive cellular therapy[2][6][7][8]. The mechanism involves co-stimulation and proliferative activation of T cell- and antibody-mediated immunity, with evidence for both local and systemic immune responses. The primary indication is experimental immunotherapy for certain cancers, notably leukemia and melanoma.
