Overview

CD47-CAR-T cells (B6H12 scFv) is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target CD47, a cell surface glycoprotein that functions as a "don't eat me" signal by binding to SIRPα on macrophages. Developed through a collaboration involving Bioceltech, the Torrey Pines Institute for Molecular Studies, and Vietnamese research institutions, this therapy utilizes a CAR construct incorporating a single-chain variable fragment (scFv) derived from the B6H12 mouse monoclonal antibody. CD47 is frequently overexpressed in various hematological and solid tumors, contributing to immune evasion, metastasis, and angiogenesis. By redirecting T-cells to recognize and eliminate CD47-expressing cells, the therapy aims to bypass tumor immune suppression and induce direct cytotoxic killing. Preclinical studies have demonstrated efficacy against pancreatic, ovarian, and cervical cancer cell lines, as well as in xenograft models. A significant technical challenge for this approach is T-cell fratricide, as T-cells themselves express CD47, necessitating strategies to prevent self-destruction of the therapeutic cells.
