CD5 CAR T-cells (Zhejiang University) is an investigational chimeric antigen receptor (CAR) T-cell therapy being developed for the treatment of relapsed or refractory CD5-positive T-cell acute lymphoblastic leukemia (T-ALL). The therapy involves the genetic modification of T cells to express a CAR that recognizes the CD5 antigen, a 67-kDa transmembrane glycoprotein expressed on the surface of nearly all mature T cells and most T-cell malignancies. To overcome the challenge of "fratricide"—the self-destruction of CAR-T cells that also express the CD5 target—the manufacturing process typically involves the deletion of the endogenous CD5 gene using CRISPR/Cas9 or similar gene-editing technologies. The engineered cells are designed to selectively bind to and lyse CD5-positive leukemic blasts upon intravenous infusion.
