Overview

This investigational multi-peptide cancer vaccine is a pentavalent biologic designed to target five cancer-testis antigens: CDCA1 (cell division cycle associated 1), URLC10 (up-regulated in lung cancer 10), KIF20A (kinesin family member 20A), DEPDC1 (DEP domain containing 1), and MPHOSPH1 (M-phase phosphoprotein 1). Developed by Tokyo Medical University in collaboration with St. Luke's International Hospital and the University of Tokyo, the vaccine consists of synthetic HLA-A*24:02-restricted peptides. These antigens are ideal targets for immunotherapy as they are overexpressed in various cancers but have limited expression in normal tissues, except for immune-privileged sites like the testis. The vaccine is emulsified with the adjuvant Montanide ISA 51 and administered subcutaneously to induce cytotoxic T lymphocyte (CTL) responses against tumor cells expressing these specific antigens. It has been evaluated in Phase I clinical trials for HLA-A*24:02-positive patients with refractory metastatic breast cancer.
