Overview

CDH3 ADC (duocarmycin) is a preclinical antibody-drug conjugate (ADC) developed by Nerviano Medical Sciences (NMSCMS). It is designed to target P-cadherin (CDH3), a cell-surface protein that is frequently overexpressed in various epithelial cancers, including breast, lung, and pancreatic tumors, while showing limited expression in normal tissues. The ADC consists of a monoclonal antibody specific for CDH3 conjugated to a novel duocarmycin-based payload. Duocarmycins are potent cytotoxic agents that act as DNA alkylators, binding to the minor groove of DNA and causing irreversible alkylation that leads to cell death. By selectively delivering this payload to CDH3-expressing tumor cells, the ADC aims to maximize therapeutic efficacy while minimizing systemic toxicity. The program is currently in the preclinical stage of development.
