Overview

CDX‑1401 + resiquimod + poly‑ICLC is a combination immunotherapy regimen under investigation for the treatment of cancers expressing the NY‑ESO‑1 tumor antigen. CDX‐1401 is a fully human monoclonal antibody targeting the dendritic cell receptor DEC‐205, genetically fused to the full-length NY‐ESO‐1 tumor antigen, designed to deliver this antigen directly to dendritic cells and induce robust immune responses against cancer cells expressing NY‑ESO‑1[1][6]. Resiquimod is a Toll-like receptor 7/8 agonist that acts as an immune adjuvant, while poly–ICLC (Hiltonol) is a synthetic double-stranded RNA acting as a Toll-like receptor 3 agonist and further stimulates innate immunity[4][7]. The combination aims to enhance both humoral and cellular immunity against tumors. Developed primarily by Celldex Therapeutics, this regimen has been evaluated in phase 1 and phase 2 clinical trials for advanced malignancies including melanoma and other solid tumors[2][4].
