CEB-01 + SN-38 is a combination therapy consisting of a novel biocompatible and biodegradable membrane implant (CEB-01) made from poly lactic-co-glycolic acid (PLGA) nanofibers entangled with the cytotoxic agent SN-38. This implant is designed for loco-regional, steady, and prolonged release of SN-38 directly into the surgical bed after resection of retroperitoneal sarcomas. The goal is to increase local control rates and survival by maintaining therapeutic concentrations of SN-38 at the tumor site while minimizing systemic exposure. SN-38 is the active metabolite of irinotecan, a topoisomerase I inhibitor, and exhibits 100–2000 times greater cytotoxicity than irinotecan itself. However, due to its poor solubility and instability at physiological pH, direct clinical use has been limited; thus, delivery systems like CEB-01 are being developed to overcome these challenges[1][3][5]. The combination has shown promising results in early-phase clinical trials for preventing local recurrence in retroperitoneal sarcoma patients following surgery[1][2][4].
