Overview

Cediranib + cilengitide is a combination therapy that was investigated for the treatment of recurrent glioblastoma. Cediranib is a vascular endothelial growth factor receptor (VEGFR) tyrosine kinase inhibitor that works by blocking blood flow to tumors. Cilengitide is an integrin inhibitor. The combination was designed to target both angiogenesis and tumor invasion pathways, as integrins are upregulated after VEGF inhibition and may play a role in resistance mechanisms to anti-VEGF therapy. Despite showing pharmacodynamic changes in blood and imaging biomarkers, the combination did not demonstrate sufficient survival and response rates to warrant further development.
