Overview

Cefamandole + gentamicin is a combination of two antibiotics used primarily for the treatment of severe bacterial infections, especially those caused by gram-negative organisms. Cefamandole is a second-generation cephalosporin (beta-lactam) antibiotic that acts by inhibiting bacterial cell wall synthesis through binding to penicillin-binding proteins, leading to cell lysis and death. Gentamicin is an aminoglycoside antibiotic that binds to the 30S subunit of bacterial ribosomes, inhibiting protein synthesis and resulting in bactericidal activity. The combination exploits synergistic effects between beta-lactams and aminoglycosides, enhancing efficacy against certain pathogens and reducing relapse rates compared to monotherapy. This regimen has been used successfully in conditions such as pyelonephritis, septicemia, respiratory tract infections, urinary tract infections, skin/soft tissue infections, bone/joint infections, and peritonitis[1][2][6][7][9].
