cefazolin + piperacillin + tazobactam is a combination antibiotic regimen investigated for the prevention of postoperative infections in patients with severe burns. Cefazolin is a first-generation cephalosporin that provides coverage against Gram-positive cocci, including methicillin-susceptible Staphylococcus aureus (MSSA), by inhibiting bacterial cell wall synthesis through the inactivation of penicillin-binding proteins (PBPs). Piperacillin is an extended-spectrum ureidopenicillin that targets a wide range of Gram-positive and Gram-negative bacteria, as well as anaerobes, also by inhibiting PBPs. Tazobactam is a penicillanic acid sulfone beta-lactamase inhibitor that protects piperacillin from degradation by Class A and Class C beta-lactamases. This combination or the comparative use of these agents has been primarily studied by Assistance Publique - Hôpitaux de Paris in the Phase 3 PROBURN trial to optimize prophylactic antibiotic strategies in the high-risk burn population.
