Drug intelligence / Profile preview
This refers to the antibiotic regimens evaluated in the **ANTEIPA** clinical trial (NCT03142204), sponsored by the **Centre Hospitalier Intercommunal Creteil**. The trial is a Phase 2 randomized non-inferiority study comparing two treatment strategies for the eradication of early *Pseudomonas aeruginosa* (PA) infection in adult patients with bronchiectasis. The experimental "monotherapy" regimen consists of oral **ciprofloxacin** (a fluoroquinolone) combined with nebulized **colistimethate sodium** (a polymyxin). This is compared against a "bitherapy" regimen that adds intravenous **ceftazidime** (a third-generation cephalosporin) to the ciprofloxacin and colistimethate sodium combination. Ciprofloxacin inhibits bacterial DNA replication by targeting DNA gyrase and topoisomerase IV. Colistimethate sodium acts as a surfactant that disrupts the bacterial outer membrane by binding to lipopolysaccharides. Ceftazidime inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins. The study aims to determine if the simpler, non-intravenous-containing regimen is as effective as the standard intensive regimen in eradicating early PA infection and preventing chronic colonization.
