Overview

Ceftriaxone + clarithromycin is a combination of two antibiotics used primarily for the treatment of community-acquired pneumonia (CAP) and other bacterial infections. Ceftriaxone is a third-generation cephalosporin that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins, leading to cell lysis and death. Clarithromycin is a macrolide antibiotic that inhibits bacterial protein synthesis by binding to the 23S rRNA of the 50S ribosomal subunit, blocking translocation during translation. The combination has been shown to be more effective than ceftriaxone alone in achieving higher cure rates and faster clinical stability in pediatric CAP without increasing adverse events[1][2]. Clarithromycin also reduces pneumolysin production, an important virulence factor in Streptococcus pneumoniae, even among macrolide-resistant strains[2][3].
