Celastrol-conjugated chitosan oligosaccharide (Cel-CSO) is a polymer-drug conjugate designed to improve the therapeutic profile of celastrol, a pentacyclic triterpene derived from Tripterygium wilfordii. By covalently attaching the hydrophobic celastrol to a water-soluble chitosan oligosaccharide (CSO) backbone via an amide bond, the conjugate achieves significantly higher aqueous solubility (18.6 mg/mL) and prolonged systemic circulation. In preclinical models of pancreatic cancer (BxPC-3 cells), Cel-CSO demonstrated potent anti-tumor activity, induction of apoptosis, and suppression of metastasis, while exhibiting lower hepatotoxicity and subacute toxicity compared to free celastrol.
