Celecoxib + calcitriol is an experimental combination therapy consisting of two active pharmaceutical ingredients: celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, and calcitriol, the hormonally active form of vitamin D3. Celecoxib acts as a nonsteroidal anti-inflammatory drug (NSAID) that inhibits COX-2 to reduce inflammation and pain. Calcitriol regulates calcium homeostasis and exhibits antiproliferative effects in various cancer cell lines. Preclinical studies have demonstrated that the combination of celecoxib and calcitriol produces synergistic or additive growth-inhibitory effects in breast and ovarian cancer cell lines by inhibiting proliferation more effectively than either agent alone[1][2][5]. The mechanism involves both direct inhibition of tumor cell growth by vitamin D signaling through the vitamin D receptor (VDR), as well as suppression of prostaglandin E2 synthesis via COX-2 inhibition by celecoxib.
