celecoxib + durvalumab + tremelimumab is an experimental combination therapy being investigated for the treatment of advanced or metastatic hepatocellular carcinoma (HCC). This regimen is the focus of the Phase II RECON study (NCT05741151), led by Emory University. It combines celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, with the immune checkpoint inhibitors durvalumab (an anti-PD-L1 monoclonal antibody) and tremelimumab (an anti-CTLA-4 monoclonal antibody). The rationale for this combination is that COX-2 inhibition by celecoxib may reduce the production of immunosuppressive prostaglandins in the tumor microenvironment, potentially overcoming resistance to the dual checkpoint blockade provided by durvalumab and tremelimumab.
