Overview

Celecoxib + eflornithine is an investigational oral combination therapy consisting of celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, and eflornithine (also known as difluoromethylornithine or DFMO), an irreversible inhibitor of ornithine decarboxylase. This combination has been studied primarily for the chemoprevention and management of familial adenomatous polyposis (FAP), a hereditary condition characterized by the development of numerous colorectal adenomas with high risk for progression to colorectal cancer. The rationale for combining these agents is based on their complementary mechanisms—celecoxib reduces prostaglandin-mediated inflammation and polyp growth via COX-2 inhibition, while eflornithine suppresses polyamine synthesis, which is implicated in cell proliferation and tumorigenesis. Clinical trials have shown that this combination can reduce global polyp burden more effectively than celecoxib alone in FAP patients[1][3]. The safety profile appears acceptable with no significant increase in adverse events compared to monotherapy[1][3].
