Celecoxib + gefitinib is a combination of two small molecule drugs used experimentally in oncology. Celecoxib is a selective cyclooxygenase-2 (COX-2) inhibitor, while gefitinib is an epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor. This combination has been studied for its potential to enhance antitumor effects by simultaneously inhibiting COX-2-mediated inflammation and EGFR-driven cell proliferation and survival pathways. Early-phase clinical studies have shown that the combination is well-tolerated and may provide enhanced efficacy compared to either agent alone, particularly in cancers such as squamous cell carcinoma of the head and neck (SCCHN) and docetaxel-resistant prostate cancer[1][2]. The rationale for combining these agents lies in their complementary mechanisms—celecoxib reduces prostaglandin synthesis involved in tumorigenesis, while gefitinib blocks EGFR signaling critical for cancer cell growth.
