Overview

This is an investigational combination therapy consisting of four agents—celecoxib, interferon alpha-2b, pembrolizumab, and rintatolimod—being studied for the treatment of metastatic or unresectable triple negative breast cancer. Each component has a distinct mechanism of action: - Celecoxib is a selective cyclooxygenase-2 (COX-2) inhibitor that reduces prostaglandin synthesis and inflammation. - Interferon alpha-2b is a recombinant cytokine with immunomodulatory and antiproliferative effects. - Pembrolizumab is a monoclonal antibody targeting programmed cell death protein 1 (PD-1), acting as an immune checkpoint inhibitor to enhance T-cell mediated antitumor activity. - Rintatolimod is a synthetic double-stranded RNA that acts as a Toll-like receptor 3 (TLR3) agonist, stimulating innate immune responses. The rationale for this combination centers on synergistically enhancing both innate and adaptive antitumor immunity by combining COX-2 inhibition, cytokine therapy, checkpoint blockade, and TLR3 activation. This regimen is currently under clinical investigation in patients with advanced triple negative breast cancer[1].
