Celecoxib + metformin is a drug combination that has shown synergistic effects in inhibiting tumor growth, particularly in hepatocellular carcinoma (HCC) and non-small cell lung cancer (NSCLC). The combination works by reducing the phosphorylation of mTOR and causing reactive oxygen species (ROS) aggregation in cancer cells, leading to DNA double-strand breaks and increased expression of the tumor suppressor factor p53. This combination has demonstrated superior efficacy compared to either drug alone in both in vitro and in vivo studies, showing enhanced inhibition of cell proliferation, migration, and increased induction of apoptosis. The combination therapy also appears to suppress Raf-MEK-ERK cascades and PI3K-AKT signaling, which may help overcome drug resistance.
