Overview

This combination therapy consists of the PD-1 monoclonal antibody cemiplimab, the BRAF inhibitor dabrafenib, and the MEK inhibitor trametinib. It is being investigated for the treatment of BRAF-V600E mutant anaplastic thyroid cancer (ATC), particularly in patients who have progressed on prior BRAF/MEK inhibitor therapy. The rationale is that BRAF and MEK inhibition may enhance the tumor's sensitivity to immune checkpoint blockade by modulating the tumor microenvironment and increasing antigen expression. Cemiplimab acts as an immune checkpoint inhibitor by blocking the PD-1 receptor on T-cells, while dabrafenib and trametinib target the MAPK signaling pathway to inhibit tumor cell proliferation.
