Cemiplimab is a fully human monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, functioning as an immune checkpoint inhibitor to enhance T-cell mediated anti-tumor responses. It is primarily developed for and approved in certain skin cancers but is being investigated in combination with chemotherapy for other malignancies, including pancreatic cancer. Gemcitabine is a nucleoside analog antimetabolite chemotherapy agent that inhibits DNA synthesis by incorporation into DNA and inhibition of ribonucleotide reductase, leading to cell death. The combination of cemiplimab and gemcitabine leverages both immunotherapy and cytotoxic mechanisms to treat advanced cancers such as metastatic pancreatic ductal adenocarcinoma[1][3][6][8].
