This is a combination therapy comprising **cemiplimab** (a monoclonal antibody against PD-1), **ipilimumab** (a monoclonal antibody against CTLA-4), and **chemotherapy** (typically a platinum-based doublet such as carboplatin or cisplatin plus another agent). Cemiplimab works by blocking programmed death receptor-1 (PD-1), thereby releasing inhibition of the immune response against cancer cells. Ipilimumab blocks cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), also checkpoint inhibition, further promoting immune-mediated anti-tumor activity. The chemotherapy component (such as carboplatin/paclitaxel, carboplatin/pemetrexed) works by directly damaging cancer cell DNA or interfering with cell division, leading to cytotoxicity. Combination approaches with two immune checkpoint inhibitors and chemotherapy are under clinical investigation for the treatment of advanced cancers, including non-small cell lung cancer and melanoma, to enhance efficacy by attacking tumors via complementary immune-activating and cytotoxic mechanisms[2][3][5].
