Drug intelligence / Profile preview
cemsidomide + dexamethasone + elranatamab (Elrexfio) is an investigational combination regimen being developed for the treatment of relapsed/refractory multiple myeloma (RRMM). The regimen combines cemsidomide (CFT7455), an oral small-molecule molecular glue degrader (MonoDAC) that targets the transcription factors IKZF1 (Ikaros) and IKZF3 (Aiolos) via cereblon (CRBN) E3 ligase modulation; dexamethasone, a corticosteroid; and elranatamab (Elrexfio), an FDA-approved bispecific T-cell engager (TCE) targeting B-cell maturation antigen (BCMA) on myeloma cells and CD3 on T cells. This combination is designed to synergistically enhance anti-myeloma efficacy by combining direct tumor cell degradation and robust T-cell activation with targeted T-cell redirection. The regimen is being evaluated in a Phase 1b clinical trial (NCT07280013) sponsored by C4 Therapeutics, with Pfizer supplying elranatamab.
